How do I move the roles and other configuration from Dev to QA and Prod when I make changes in Dev?
Do I have to do it manually?

A good way of moving the roles and other configuration information is using the Management REST API, which allows you to import/export roles. This is covered in the Scripted Administration in the product doc.
